Identification of genetic evidence for dobrava virus spillover in rodents by nested reverse transcription (RT)-PCR and TaqMan RT-PCR.

A survey of 158 rodents caught in the Czech Republic identified Dobrava virus sequences closely related to that of the Dobrava virus type strain in Apodemus sylvaticus and Mus musculus rodents. The identity of A. sylvaticus was unequivocally confirmed by random amplified polymorphic DNA analysis. The data seem to indicate hantavirus spillover from Apodemus flavicollis to other rodents.

Identification of spring viraemia of carp virus (SVCV) by combined RT-PCR and nested PCR.

A combination of single-tube reverse transcription (RT)-PCR and nested PCR was used to identify spring viraemia of carp rhabdovirus (SVCV) in infected cell cultures and fish tissues. Two pairs of specific primers (external and internal) were selected from the glycoprotein gene sequence. A specific product of 470 bp was amplified from RNA derived from 34 SVCV isolates including the reference str...

Nested-RT-PCR and multiplex RT-PCR for diagnosis of rhinovirus infection in clinical samples.

Human rhinoviruses (HRVs) are positive-stranded RNA viruses belonging to the Enterovirus genus in the family of Picornaviridae. Identification of the specific strain in HRV disease has been difficult because the traditional serological method is insensitive, labor intensive, and cumbersome. With the fast progress in molecular biological technique, more sensitive and faster molecular methods hav...

Use of elevated reverse transcription reaction temperatures in RT-PCR.

The use of reverse transcriptase coupled with the polymerase chain reaction (RT-PCR) has been proven to be a valuable tool in the synthesis and isolation of cDNA clones as well as the quantification of rare messenger RNA species. While a number of different approaches can be used to permit gene-specific cDNA synthesis and amplification, a recurring problem is one of high background signal (i.e....
